چکیده

To determine cholesterolin biologicalfluids, use of the ferric chloride-sulfuric acid reagent introduced by Zlatkis et al. (1) is relatively simple and reliable (2, 3), and the sensitivityof the reagent for cholesterol is four to five times greater than that of the Liebermann-Burchard reagent (2). However, Parekh and Jung (4) reported that hydrogen chloride is evolved during the ferric chloride-sulfuric acid reaction, leading to poor control of reaction temperature. In addition, the very deliquescent ferric chloride is tedious to handle. The ferricchloride-sulfuricacid method would thus be better if a nondeliquescent iron saltcould be substituted that forms no volatile compounds on reaction with sulfuric acid. We studied the possibility of using ferric ammonium sulfate.

Simplified determination of the lipid components of blood serum.

A simplified scheme of analysis is presented for the determination of free and esterified cholesterol, the phospholipids, and the neutral fats (triglycerides) of blood serum. The hydroxamic acid reaction is used to determine total esterified fatty acids. The phospholipids are determined after acid digestion of a trichloroacetic acid precipitate of protein-phospholipid, and both total and free c...

مقایسه روش‌های پروتئین‌زدایی در سنجش نیتریک اکساید سرمی به روش گریس

Background: Nitric oxide (NO) is involved in numerous physiologic and phathophysiologic processes. Recently, further investigators have focused on serum NO determination. In our previous study, we validated a simple, cheap and rapid method for serum NO determination based on the Greiss reaction. Deproteinization is a necessary step for this reaction, thus, the present study was designed to asse...

Determination of Urea in Blood and Urine with Diacetyl Monoxime-glucuronolactone Reagent.

N UMEROUS STUDIES have been reported in the literature on the photometric determination of urea in biological fluids with diacetyl monoxime as the color-developing agent. The reaction is photosensitive, does not follow Beer’s law, and requires special precautions for reproducible results. In many proposed procedures, oxidants were added in the reaction mixture to destroy hydroxylamine which mig...

New cholesterol reagent.

R ECENTLY a new reagent was proposed for the determination of cholesterol (1, 2). This reagent was purported to be saturated ferrous sulfate in glacial acetic acid. Since it has been reported that ferrous sulfate is qualitatively insoluble in anhydrous acetic acid (3, 4) and that ferrous iron is “subject to pronounced air oxidation in glacial acetic acid” (5), it was felt that several factors s...
